Calculate the MFI (Mean/Median Fluorescent Intensity) by using the Export Statistics Task.
1. Add a Export Statistics task to your workflow
Click Add new child task and select Export Stats from the task selector. Note that your Workflow might look slightly different, if you are using Compact View.
You can also access the Export Statistics task from within other tasks, such as the Gating task. Have a look below the Logout button.
2. Setup your Export Statistics task
2.1. Select your Files
Select the Files you want to export MFI for and click Next or click Features.
Most dropdowns in OMIQ allow you to filter the list, just start typing. You can even filter by metadata.
3. Select your Features
Select the Filters you want to export MFI for → select the Mean Fluorescent Intensity and/or the Median Fluorescent Intensity in Feature Measures → select the Features you want the MFI for. You can either click Next to add more data to your Export Statistics task, or click Configuration if you only wish to export the data shown in this step.
In this example, we will export the Mean and Median Fluorescent Intensities for CCR5, CCR7 and CCR6 on CD4+, CD8+, CD4+CD8+ and DN cells.
The Mean Fluorescent Intensity may be most useful when used to describe normal distributions whereas the Median Fluorescent Intensity is a better indicator of the central tendency of your data and therefore a more useful measure for bi-modal or multi-modal data. Additionally, the Arithmetic Mean is affected by outliers, while the Median is less sensitive to outliers.
4. Configure your Export settings
Select the File Format you want to export to, either as a CSV file or a GraphPad Prism file. Adjust any other settings and click Export or Run Export Stats.
Change the Exported Data Units to Scaled per Scaling Settings if you prefer to report any scaling you have applied in workflow.
5. Download your File
Navigate the the Results tab within the Export Statistics task and click the cloud icon to download your file.
